Workshop Summary

This program will focus on novel science, insights, tools, and approaches for targeting and bypassing the CNS barriers to achieve drug delivery to the brain and spinal cord. Specialized barriers between the CNS and the bloodstream include the blood-brain barrier and the blood-cerebrospinal fluid barriers, among others. Highly limited paracellular transport and the expression of numerous efflux transporters at these key endothelial and epithelial interfaces pose major challenges to successful brain delivery of both small and large molecule therapeutics.

The goal of this workshop is to both clarify the main issues for the field as well as discuss the most cutting edge current and future strategies for CNS drug delivery. The program will bring together international experts and junior investigators from both academic, government (i.e., regulatory),and industry environments and provide a format promoting exchange of new ideas and brainstorming novel solutions to existing challenges. The workshop will also serve to bring together members and stakeholders from the American Association of Pharmaceutical Scientists (AAPS) and the International Brain Barriers Society (IBBS) to foster future collaboration.

Specific themes for the workshop will cover the following topics:physiology and regulation of the CNS barriers, molecular mechanisms of brain permeability and targeting, novel CNS targeting strategies for crossing and bypassing the barriers, models, and in vitro/ in vivo correlations. Progress and challenges in drug discovery and development along with the use of cutting-edge methodologies will also be addressed.

Participants of this will:

  1. Review the physiology and regulation of brain barriers
  2. Explain the various molecular mechanisms involved in brain barriers permeability of small and large therapeutics i.e., specific membrane transporters and receptors
  3. Understand novel approaches for brain barriers drug targeting and delivery
  4. Discuss cutting edge in vitro and in vivo methodologies for the study of drug delivery across brain barriers
  5. Address various novel approaches for drug delivery to the brain by bypassing or crossing the brain barriers
  6. Present and discuss: IVIV correlations and PBPK models in brain drug disposition
  7. Summarize the progress and remaining challenges in the field of CNS drug targeting and further discuss with attendees in a panel session
  8. Discuss with experts from academia, government, biotech and big pharma together to come to an understanding of how to lead the field efficiently forward
  9. Analyze specific cases addressing challenges faced in the field of CNS drug discovery and development in the industry in round table format
